Cam timing versus crank timing---cam swap

Gents I'm in the middle of a cam swap and have an important question before I install the pushrods and button it up. Bike is an 09 Big Twin.

WHen measuring the cam and crank sprockets offset, I turned the crank in order to get measurements across the sprocket faces in several differenct positions for the correct cam spacer, so now the crank sprocket is no longer in the position it was when I pulled off the primary cam/crank sprockets chain as it was before I started the job.

My question is-- is it possible to get the crank timing 180 degrees out from the cam timing even though the sprocket timing marks are lined up?

Or, as long as the marks are in line, no matter where the crank stopped after I rotated it will the cam/crank timing be right?

Thanks for your time with this one-- trying to finish it up today all going well.

All good so far--rotated engine in 6th gear after adjustable pushrods bled down--- all seems fine and the cam side is buttoned up. In addition to your help I could also see when I was on the base circle of each cam, as each exhaust lifter rolled over the 'decompression trigger' on the EZ start cams.

I'll get the cat gutted while waiting on the ECM to come back from RevPerformance--then can hold my breath when I hit the start button. I must admit I scratched my head far more times than I thought I would during the cam install. Still a week or so away from firing up.

Thanks-- I also understand it isn't possible to be 180 out with a single pin crank.... I hate those brain farts when they happen-- just couldn't "see it" before help arrived LoL! Freakin stood there reading the SM and the cam install destructions over and over and just couldn't get it in my head...
